A phylogeographic study of Polygonatum in the Southeastern United States using AFLPs, DNA sequences, and morphological data.

Polygonatum (Asparagaceae) comprises cosmopolitan, geophytic, woodland plants found throughout North America, Europe, and Asia. Two species are currently recognized in the Southeastern United States: Polygonatum biflorum (Walter) Elliott and P. pubescens (Willdenow) Pursh. These two species are similar in appearance but can be distinguished on the basis of several morphological characters. The most obvious of these distinctions is that P. pubescens has leaves that are pubescent on the abaxial surface whereas the leaves of P. biflorum are glabrous. Polygonatum biflorum (sensu lato), which includes P. biflorum var. commutatum and P. biflorum var. hebetifolium, is a taxonomically difficult polyploid complex characterized by plants that vary in size and ecology as well as reproductive morphology. As such, this group is in need of taxonomic revision and further morphological and molecular analysis. The current study employs phylogeographic methods to test current taxonomy and to investigate the origin of polyploidy in the P. biflorum species complex. This will be accomplished through a comparative analysis of genetic and morphological variation among field collected specimens from populations of P. biflorum and P. pubescens ranging from Ohio to Alabama, using AFLPs, flow cytometry, DNA sequence data,and a morphological character matrix. Herbarium specimens will be used to expand the survey of morphological variation throughout the native range of Polygonatum in North America. A principle components analysis will be used to examine morphological variation in the context of taxon delimitation and phylogeography. Resolution of hypotheses of evolutionary history of Polygonatum in the Southeast will be discussed along with their implications for taxonomic revisions.
