Establishment of an in vitro protocol of Chuquiragua (Chuquiraga jussieui J.F. Gmel) from apical and axillary buds.

The establishment of native and vulnerable plant species in in vitro culture has become a major research objective, with the goal of conserving plants through the standardization of micropropagation protocols. Chuquiragua (Chuquiraga jussieui J.F. Gmel.) an Andes mountains shrub genus, is an ideal candidate for the development of these cultures. This species is known locally as Chuquirahua, Walker flower, and Andes´s flower. Through the years it has been threatened by poor utilization of soils, as well as reforestation with species that erode the land, leading to the reduction of Chuquiragua in its native habitat. In this project, shoots of apical and axillary buds were induced on MS 50% medium (1962) in the presence of BAP (0.1 to 2.0 mg/L), IBA (0.0 - 0.1 mg/L) and BRA (2.0 mg/L) for 30 days. Then, generated buds were transferred to a MS 50% multiplication medium, in the presence of BAP (0.1 to 2.0 mg/L), IBA (0.0 - 0.1 mg/L) and BRA (5 mg/L), followed by incubation for 30 days. Subsequently, obtained buds were induced to rooting in MS 50% medium in the presence of IBA (2.0 - 6.0 mg/L) and BAP (0.0 - 0.1 mg/L). The results reflected that MS 50% medium supplemented with: BAP 2.0 mg/L + BRA 2 mg/L, allows the shoot induction of apical and axillary buds in a 92%, and  also MS 50% medium supplemented with: BAP 2.0 mg/L + BRA 5 mg/L during multiplication phase, permits obtain largest number of buds. To finish, 70% of buds from roots were obtained with MS 50% medium supplemented with: IBA 6.0 mg/L BAP + 0.1 mg/L. The micropropagation of Chuquiragua is critical to conservation efforts in Andean ecosystems due to the important ecological role that it plays.
